48-week obesity trial

In a randomized trial of 338 adults, the 12 mg study arm had a 24.2% least-squares mean reduction in body weight at 48 weeks, compared with 2.1% for placebo. The study’s primary endpoint was measured at 24 weeks; the 48-week result was a secondary endpoint.

Gastrointestinal adverse events were common, and dose-dependent increases in heart rate were observed. Study-arm amounts describe the trial, not product-use instructions. Funded by Eli Lilly.

80-week obesity trial

TRIUMPH-1 randomized 2,339 adults with obesity without diabetes. At 80 weeks, the reported mean body-weight change was −25.0% in the 12 mg arm and −3.9% with placebo, using a treatment-regimen (intention-to-treat) estimand.

Gastrointestinal events were the most common adverse events. Population, trial duration, and analysis differ from the phase 2 trial; these results should not be compared as equivalent estimates. Funded by Eli Lilly.
